What This Study Did, in Plain Language

What Was Tested

This registered phase 4 trial tested Exenatide.

Adipose tissues, which include white adipose tissue (WAT) and brown adipose tissue (BAT), play an essential role in regulating whole-body energy homeostasis.

Until 2009 the question of whether adult humans had BAT and whether it could conceivably contribute to whole body energy usage in a meaningful way was a matter of vigorous debate.

It had two arms (groups): GLP-1 agonism group (experimental) and Placebo group (placebo comparator).

The registry classes the interventions as drug.

More on the Plan

Individuals were treated for 10 days.

Biopsy for subcutaneous white adipose (1.5X1.5X1.5cm) was performed before and after 10 days treatment programme under local anesthesia.

Measure the brown fat characteristics of biopsy samples. The sample was immediately processed in 3 sections. One part was stored for immunohistology and western blot, the second was snap-frozen for estimation of biochemical markers, and the remainder was used to harvest small subcutaneous arteries with micro-dissection.

Who Took Part

The registry lists 20 actual participants took part.

It was open to men only aged from 20 years up to 30 years.

Healthy volunteers were not accepted; participants needed the condition being studied.

Examples of who could join: Body mass index (BMI) \> 30 kg/m2. Age 20 - 30 years old.

The full record lists 3 inclusion and 4 exclusion criteria; among the exclusions: BMI \< 30 kg/m2; use of medicines.

How the Study Was Run

It was run as a randomized, parallel-group and open-label study.

Random assignment meant chance, not the researchers, decided who got Exenatide and who got the comparison, which limits bias.

The Exenatide group and the comparison group ran side by side rather than switching.

Everyone knew when Exenatide was being given (open label), which is simpler to run but leaves more room for expectations to affect what is reported.

It started in 2014-06 with completion listed for 2015-07.

It ran at 1 site in China.

Its stated primary purpose is treatment.

What Was Measured

Researchers chose this as the main measure: The expression of brown adipose related genes in subcutaneous white adipose tissue (time frame: 3 months).

What This Means in Plain Language

Put simply, this is a phase 4, randomized trial of Exenatide in 20 people.

Its size is small, so on its own it can only give an early look.

The trial is marked completed; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound

Exenatide: A 39-amino-acid GLP-1 receptor agonist first identified in Gila monster venom; sold as Byetta and Bydureon for type 2 diabetes.
