What This Study Did, in Plain Language

What Was Tested

In this not phased trial, researchers set out to test NAD+ for insulin resistance, prediabetes & type 2 diabetes.

Nicotinamide riboside is a dietary supplement which is similar to vitamin B3.

It had two arms (groups): Arm I (home exercise) (active comparator) and Arm II (home exercise, nicotinamide riboside) (experimental).

Patients complete 18 home exercise sessions over 30 minutes each, 3 days per weeks for 6 weeks.

Patients complete home exercise as in Arm I. Patients also receive nicotinamide riboside PO daily for 6 weeks.

The registry classes the interventions as dietary supplement and other.

More on the Plan

Describe the effect of exercise with or without nicotinamide riboside (NR) on hyperglycemia and skeletal muscle health in CCS with a history of prediabetes.

OUTLINE: Patients are randomized to 1 of 2 arms.

ARM I: Patients complete 18 home exercise sessions over 30 minutes each, 3 days per weeks for 6 weeks.

ARM II: Patients complete home exercise as in Arm I.

Who Took Part

The registry lists 20 actual participants took part.

It was open to people of any sex aged from 18 years with no upper age limit.

Healthy volunteers were not accepted; participants needed the condition being studied.

Examples of who could join: In remission at time of enrollment. At least 18 years of age at time of enrollment. Able to access online exercise program at home. Ability to tolerate the prescribed resistance exercise program. Able to understand and sign the study specific informed consent form (ICF).

The full record lists 9 inclusion and 6 exclusion criteria; among the exclusions: taking a nicotinamide adenine dinucleotide (nad)+ precursor in the two weeks prior to enrollment; currently taking medication for hyperglycemia or diabetes; females who are pregnant or planning to become pregnant.

How the Study Was Run

It was run as a randomized, parallel-group and open-label study.

Because people in this NAD+ study were split into groups at random, differences between the groups are less likely to reflect who happened to join each one.

The NAD+ group and the comparison group ran side by side rather than switching.

Nothing was hidden in this open-label NAD+ study: the insulin resistance, prediabetes & type 2 diabetes participants and staff both knew the treatment.

It started in 2022-06-23 with completion listed for 2026-09-21.

It ran at 1 site in United States.

Its stated primary purpose is supportive care.

What This Means in Plain Language

Put simply, this is a not phased, randomized trial of NAD+ in 20 people with insulin resistance, prediabetes & type 2 diabetes.

Its size is small, so on its own it can only give an early look.

Its status is active not recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound

NAD+: The coenzyme that accepts electrons at complex I.
