What This Study Did, in Plain Language
What Was Tested
In this not phased trial, researchers set out to test Therapeutic nicotine (patch, non-peptide).
People with major depressive disorder (MDD) are twice as likely to use nicotine compared to the general population.
They have greater withdrawal symptoms and are more likely to relapse after quitting compared with smokers without MDD.
It had two arms (groups): Arm 1: Placebo (placebo comparator) and Arm 2: Nicotine Patch (experimental).
Placebo patch + Placebo Pill.
Nicotine Patch + Placebo Pill.
The registry classes the interventions as drug and other.
More on the Plan
Individuals with major depressive disorder (MDD) are more vulnerable for experiencing these burdens as they are twice as likely to use nicotine versus the general population.
The current work will explain the neurobiological basis of this enhanced risk and will define potential targets for lessening the impact of nicotine on those with MDD.
This research plan will take the innovative approach of evaluating nicotine s effects in non-smokers with and without MDD.
In contrast to focusing on nicotine dependent individuals, which introduces confounds due to chronic use, this design will directly show the domains in which the neurobiological impact of nicotine is greater in those with MDD, providing a mechanistic framework for enhanced risk.
Who Took Part
The registry lists 620 planned participants were the target.
It was open to people of any sex aged from 18 years up to 60 years.
Healthy volunteers could join, not only people with the condition.
Examples of who could join: Able and willing to provide written informed consent. Both sexes and all ethnic origins, age between 18 and 60 at the time of consent. Absence of pregnancy and breastfeeding. Have a Breath Alcohol Value of 0 on all study visit days involving scanning. Participant may be rescheduled if this value is greater than 0. Meet DSM-5 diagnostic criteria for current MDD at screening Clinical judgement will be used to interpret criteria.
The full record lists 15 inclusion and 28 exclusion criteria; among the exclusions: history of moderate or severe substance use disorder in the past 6 months (other than caffeine); body mass index (bmi) lower than 18.5 kg/m\^2; contraindications to mri as determined by mri safety screening form and mock scanner trial (when available).
How the Study Was Run
It was run as a randomized, parallel-group and double-blind study.
The participants were allocated by chance, so the Therapeutic nicotine (patch, non-peptide) group and the comparison group should start out alike.
The Therapeutic nicotine (patch, non-peptide) group and the comparison group ran side by side rather than switching.
As a double-blind Therapeutic nicotine (patch, non-peptide) study, the assignments were hidden from both participants and investigators until the end.
Those kept unaware of assignments: participant and investigator.
It started in 2023-02-02 with completion listed for 2027-12-30.
It ran at 1 site in United States.
Its stated primary purpose is basic science.
What Was Measured
The main thing measured (primary outcome): 4.Nicotine effects and symptoms of MDD (time frame: At each scan visit). 3. Task and Resting State Brain (time frame: At each scan visit). 2.Task-fMRI (time frame: At each scan visit).
It also tracked 1 secondary outcome, for example Determine the relationship between blood-based biomarkers, such as inflammatory makers/metabolomics and nicotinic effects.
What This Means in Plain Language
Put simply, this is a not phased, randomized trial of Therapeutic nicotine (patch, non-peptide) in 620 people.
With 620 participants it is a large trial for this field.
It is still recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound
Therapeutic nicotine (patch, non-peptide): Pure nicotine as a signaling tool, separated from smoking and vaping.
