What This Study Did, in Plain Language
What Was Tested
The trial (not phased) is built around NAD+.
The prevalence of age-related chronic diseases (like obesity, type 2 diabetes and cardiovascular diseases) is mounting worldwide, reaching pandemic proportions.
Nicotinamide adenosine dinucleotide (NAD) is a coenzyme that regulates mitochondrial function, therefore, plays an important role in energy metabolism.
It had two arms (groups): NR+EXTR (experimental) and PLA+EXTR (placebo comparator).
This arm will ingest NR orally and perform exercise training.
This arm will ingest placebo orally and perform exercise training.
The registry classes the interventions as dietary supplement.
Who Took Part
28 planned participants were the target.
It was open to people of any sex aged from 60 years up to 80 years.
Healthy volunteers could join, not only people with the condition.
Examples of who could join: Participants are able to provide signed and dated written informed consent prior to any study specific procedures. Aged ≥ 60 and ≤ 80 years. Body mass index (BMI) 25 - 35 kg/m2. Stable dietary habits (no weight loss or gain \> 5 kg in the past 3 months). No signs of active cardiovascular disease, liver or kidney malfunction.
The full record lists 5 inclusion and 9 exclusion criteria; among the exclusions: patients with congestive heart failure and and/or severe renal and or liver insufficiency; uncontrolled hypertension; any contra-indication for mri scanning.
How the Study Was Run
It was run as a randomized, parallel-group and double-blind study.
The participants were allocated by chance, so the NAD+ group and the comparison group should start out alike.
Groups ran in parallel: each of the participants stayed in one arm throughout.
Blinding on both sides kept the participants and researchers unaware of who received NAD+.
Those kept unaware of assignments: participant and investigator.
It started in 2024-03-01 with completion listed for 2025-12.
It ran at 1 site in Chile.
Its stated primary purpose is treatment.
What Was Measured
The main thing measured (primary outcome): Skeletal muscle mitochondrial respiratory capacity (time frame: 12 weeks).
It also tracked 14 secondary outcomes, for example Quantification of proteins that regulate oxidative metabolism, Skeletal muscle mitochondrial content, Maximal aerobic capacity, Walking speed and distance, Seating and standing transitions and Exercise efficiency.
What This Means in Plain Language
Put simply, this is a not phased, randomized trial of NAD+ in 28 people.
Its size is small, so on its own it can only give an early look.
It is still recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound
NAD+: The coenzyme that accepts electrons at complex I.
