What This Study Did, in Plain Language

What Was Tested

The trial (not phased) is built around Semaglutide for fat loss, obesity & body recomposition.

This is a non-randomized, concurrent, parallel-controlled clinical trial.

The objective of this trial is to determine the relationship between weight loss responsiveness to semaglutide in obese patients and their gut microbiota.

It had two arms (groups): SLR group (sham comparator) and SHR group (active comparator).

The registry classes the interventions as drug.

More on the Plan

Obesity has gradually emerged as a major public health concern.

This project aims to investigate the individual variability in semaglutide response through multi-omics approaches including fecal metagenomic sequencing.

Utilizing in vitro bacterial screening platforms combined with gut microbiota gene knockout/heterologous expression systems, protein isolation-activity tracking, and structural characterization, the researchers will elucidate the mechanisms underlying gut microbiota-mediated semaglutide resistance from microbial, animal, and clinical perspectives.

The outcomes may identify novel therapeutic targets to overcome semaglutide resistance in weight management.

Who Took Part

Enrollment: 56 planned participants were the target.

It was open to people of any sex aged from 18 years up to 60 years.

Healthy volunteers were not accepted; participants needed the condition being studied.

Examples of who could join: Age 18-60 years. BMI ≥ 30 kg/m². At least one self-reported unsuccessful attempt at lifestyle weight loss.

The full record lists 3 inclusion and 16 exclusion criteria; among the exclusions: use of any medication for obesity indication in the past 3 months prior to screening; use of immunosuppressants, corticosteroids, antidiarrheal drugs, antibiotics, probiotics, lipid-lowering medications, and/or other gastrointestinal motility drugs in the past 3 months prior to screening; A history of endocrine-related overweight or obesity diagnoses, such as Cushing's syndrome.

How the Study Was Run

The set-up was non-randomized, parallel-group and open-label.

Groups ran in parallel: each of the fat loss, obesity & body recomposition participants stayed in one arm throughout.

Nothing was hidden in this open-label Semaglutide study: the fat loss, obesity & body recomposition participants and staff both knew the treatment.

It started in 2025-02-20 with completion listed for 2026-12-31.

Its stated primary purpose is treatment.

What Was Measured

Primary outcome: Body weight. Gut microbiota.

It also tracked 3 secondary outcomes, for example Blood biochemical indicators, The blood concentration of semaglutide and BMI.

What This Means in Plain Language

Put simply, this is a not phased, non-randomized trial of Semaglutide in 56 people with fat loss, obesity & body recomposition.

Its status is not yet recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound

Semaglutide: The active ingredient in several prescription GLP-1 receptor agonist medicines (Ozempic, Wegovy, oral tablets).
