What This Study Did, in Plain Language
What Was Tested
This registered not phased trial tested NAD+.
Adverse childhood experiences (ACEs) are directly related to cardiovascular morbidity and mortality, and impaired vascular endothelial function (VEF) is an independent predictor of future cardiovascular disease (CVD) risk \[1, 2\].
It had two arms (groups): Nicotinamide Riboside (experimental) and Placebo (placebo comparator).
Participants randomized to the control group will consume matched placebo capsules containing microcrystalline cellulose.
The registry classes the interventions as dietary supplement.
Who Took Part
30 planned participants were the target.
It was open to people of any sex aged from 18 years up to 30 years.
Healthy volunteers could join, not only people with the condition.
Examples of who could join: 18 - 30 years. ACE score of 0 OR ≥4 (Aim 1); ACE score ≥4 (Aim 2).
The full record lists 2 inclusion and 11 exclusion criteria; among the exclusions: resting arterial blood pressure \>140/90 mmhg; BMI ≥30 kg/m2 and/or weight unstable (\>2.27 kg change) last 6 month; cardiovascular, metabolic, or pulmonary disease.
How the Study Was Run
It was run as a randomized, parallel-group and triple-blind study.
Because people in this NAD+ study were split into groups at random, differences between the groups are less likely to reflect who happened to join each one.
Groups ran in parallel: each of the participants stayed in one arm throughout.
Neither the participants nor the staff measuring them knew who was on NAD+, so expectations could not easily colour the readings.
Those kept unaware of assignments: participant, investigator and outcomes assessor.
It started in 2025-03-27 with completion listed for 2026-12-31.
It ran at 2 sites in United States (2).
Its stated primary purpose is basic science.
What Was Measured
Researchers chose this as the main measure: Vascular endothelial function (time frame: Before (0 Weeks) and after 4-week supplementation period (4 Weeks)).
It also tracked 4 secondary outcomes, for example Endothelial SIRT1 Expression, Endothelial SIRT1 Activity, Cellular SIRT1 Activity in PBMCs and Cellular NAD+ metabolites.
What This Means in Plain Language
Put simply, this is a not phased, randomized trial of NAD+ in 30 people.
Its size is small, so on its own it can only give an early look.
It is still recruiting; no results are posted to the registry yet, so nothing here describes outcomes.
