What This Study Did, in Plain Language

What Was Tested

The trial (phase 2) is built around Tirzepatide.

This pilot clinical study aims to investigate the potential effects of tirzepatide on biological aging in older adults.

It had two arms (groups): Tirzepatide (active comparator) and Placebo (no intervention).

Participants will be monitored for a total of 36 weeks without receiving medication.

The registry classes the interventions as drug.

More on the Plan

Primary Objective To estimate the potential effect of tirzepatide on biological aging using epigenetic age (based on established DNA methylation-based clocks), among people 55-70 years of age with an indication for tirzepatide weight loss therapy.

The principal investigator (PI) will measure DNA methylation-based aging clocks (DNAmAge, DNAm PhenoAge, DNAm GrimAge and DunedinPACE) before and after a 24-W course of tirzepatide.

The PI will specifically assess epigenetic age acceleration (epigenetic age minus chronological age) and the pace of aging metric.

Secondary and Exploratory Objectives Among people 55-70 years of age with an indication for tirzepatide weight loss therapy, the is aim to: 1.

Who Took Part

Enrollment: 90 planned participants were the target.

It was open to people of any sex aged from 55 years up to 70 years.

Healthy volunteers were not accepted; participants needed the condition being studied.

Examples of who could join: Age 55-70 years. BMI ≥ 30 kg/m² or ≥ 27 kg/m² if also with ≥ 1 weight-related comorbidity. Willing and able to provide written informed consent and undergo all required study procedures.

The full record lists 4 inclusion and 19 exclusion criteria; among the exclusions: BMI \>35 kg/m²; uncontrolled or end-stage, comorbid, cardiometabolic disease, in the opinion of the site investigator; use of insulin.

How the Study Was Run

Design: randomized, parallel-group and double-blind.

Random assignment meant chance, not the researchers, decided who got Tirzepatide and who got the comparison, which limits bias.

Groups ran in parallel: each of the participants stayed in one arm throughout.

As a double-blind Tirzepatide study, the assignments were hidden from both participants and investigators until the end.

Those kept unaware of assignments: investigator and outcomes assessor.

It started in 2026-02-27 with completion listed for 2027-11.

It ran at 1 site in United States.

Its stated primary purpose is prevention.

What Was Measured

Researchers chose this as the main measure: Change in DNA Methylation-Based Biological Age (time frame: Immediately before and after the study visit at Weeks 4, 12, and 24).

What This Means in Plain Language

Put simply, this is a phase 2, randomized trial of Tirzepatide in 90 people.

It is still recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound

Tirzepatide: A prescription peptide medicine that acts on both the GIP and GLP-1 receptors; Mounjaro and Zepbound contain the same active ingredient.
