What This Study Did, in Plain Language

What Was Tested

The trial (phase 1) is built around Amycretin.

The purpose of this clinical study is to look at if/how much NNC0487-0111 affects food intake, appetite and metabolism in participants with obesity when compared to placebo.

There are 2 study treatments in this study.

Participants will inject the study treatment under the skin of your stomach once a week.

It had two arms (groups): NNC0487-0111 (experimental) and Placebo (placebo comparator).

Participants will receive placebo matched to NNC0487-0111 subcutaneously once weekly.

The registry classes the interventions as drug.

Who Took Part

The registry lists 120 planned participants were the target.

It was open to people of any sex aged from 18 years up to 64 years.

Healthy volunteers were not accepted; participants needed the condition being studied.

Examples of who could join: Male or female (sex at birth). Age 18-64 years (both inclusive) at the time of signing the informed consent. Considered eligible based on the medical history, physical examination, and the results of vital signs, electrocardiogram and clinical laboratory tests performed during the screening visit, as judged by the investigator. Overweight should be due to excess adipose tissue, as judged by the investigator.

How the Study Was Run

Design: randomized, parallel-group and quadruple-blind.

Because people in this Amycretin study were split into groups at random, differences between the groups are less likely to reflect who happened to join each one.

Groups ran in parallel: each of the participants stayed in one arm throughout.

Blinding on both sides kept the participants and researchers unaware of who received Amycretin.

Those kept unaware of assignments: participant, care provider, investigator and outcomes assessor.

It started in 2026-03-30 with completion listed for 2028-01-19.

It ran at 1 site in Germany.

Its stated primary purpose is treatment.

What Was Measured

The main thing measured (primary outcome): Relative change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 126 (time frame: From Baseline to Day 126). Relative change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 182 (time frame: From Baseline to Day 182). Relative change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 238 (time frame: From Baseline to Day 238).

It also tracked 17 secondary outcomes, for example Change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 126, Change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 182, Change from baseline in total energy intake across ad libitum breakfast, lunch, evening meal and snack box to Day 238, Change from baseline in total amount of food consumed across ad libitum breakfast, lunch, evening meal and snack box to Day 126, Change from baseline in total amount of food consumed across ad libitum breakfast, lunch, evening meal and snack box to Day 182 and Change from baseline in total amount of food consumed across ad libitum breakfast, lunch, evening meal and snack box to Day 238.

What This Means in Plain Language

Put simply, this is a phase 1, randomized trial of Amycretin in 120 people.

It is still recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound

Amycretin: A single molecule that activates both GLP-1 and amylin receptors, tested by injection and as a tablet in early obesity trials.
