What This Study Did, in Plain Language

What Was Tested

This registered phase 1 trial tested Eloralintide.

The main purpose of this study is to assess how well macupatide, when dosed alone and in combination with eloralintide, is tolerated and what side effects may occur in participants with obesity or overweight without Type 2 diabetes.

Blood tests will be performed to investigate how the body processes the study drug and how the study drug affects the body.

For each participant, the study will last about 4 months and will include 2 stays in the Clinical Research Unit (CRU), each lasting 3-5 nights.

It had five arms (groups): Macupatide + placebo (experimental), Eloralintide + placebo (experimental), one (experimental), one (experimental) and Placebo (placebo comparator).

Macupatide and placebo administered subcutaneously (SC).

Eloralintide and placebo administered SC.

Macupatide and Eloralintide administered SC.

The registry classes the interventions as drug.

Who Took Part

60 planned participants were the target.

It was open to people of any sex aged from 18 years up to 65 years.

Healthy volunteers could join, not only people with the condition.

Examples of who could join: Are Japanese participants assigned male at birth (AMAB) or assigned female at birth (AFAB) who agree to contraception requirements or participants who are individuals not of childbearing potential (INOCBP). Have a body mass index greater than or equal to (≥)25 kilograms per square meter (kg/m²). Have had a stable weight for the 3 months prior to screening. Stable weight is defined as less than 5 percent (%) body weight change.

The full record lists 3 inclusion and 12 exclusion criteria; among the exclusions: have known allergies to related compounds of macupatide or eloralintide, or any components of the formulations; have been diagnosed with type 1 or type 2 diabetes mellitus; have a history of chronic medical conditions involving the heart, liver, or kidneys.

How the Study Was Run

The set-up was randomized, parallel-group and double-blind.

The participants were allocated by chance, so the Eloralintide group and the comparison group should start out alike.

Groups ran in parallel: each of the participants stayed in one arm throughout.

As a double-blind Eloralintide study, the assignments were hidden from both participants and investigators until the end.

Those kept unaware of assignments: participant and investigator.

It started in 2026-10 with completion listed for 2027-06.

It ran at 3 sites in Japan (3).

Its stated primary purpose is basic science.

What Was Measured

The main thing measured (primary outcome): Number of Participants with One or More Serious Adverse Event(s) (SAEs) Considered by the Investigator to be Related to Study Drug Administration (time frame: End of Study (Week 25)).

It also tracked 5 secondary outcomes, for example PK: AUC of Macupatide in Combination with Eloralintide, PK: Maximum Concentration (Cmax) of Macupatide, PK: Cmax of Macupatide in Combination with Eloralintide and Percent Change in Body Weight.

What This Means in Plain Language

Put simply, this is a phase 1, randomized trial of Eloralintide in 60 people.

Its status is not yet recruiting; no results are posted to the registry yet, so nothing here describes outcomes.

Our Notes on the Compound
