What it is
Cagrilintide is described as an investigational amylin analog being studied for weight management. It is not a GLP-1 drug, and it is not the same thing as CagriSema, which combines cagrilintide with semaglutide.
As of October 2026, FDA is described as identifying cagrilintide as unapproved and as saying it cannot be used in compounding under federal law.
Mechanism (as described)
- Amylin is a pancreatic hormone involved in feeling full after eating. Cagrilintide was designed as a longer-acting analog to study sustained appetite and body-weight effects, and its development also addressed native amylin’s stability problems.
- Laboratory receptor studies describe it, also called AM833, as an agonist at amylin and calcitonin receptors. That differs from semaglutide’s GLP-1 receptor activity; combining the two is described as intended to engage complementary pathways.
- A second source describes a synthetic, long-acting amylin analog modified for stability and studied for activity at amylin-responsive receptor complexes formed by the calcitonin receptor and receptor activity-modifying proteins (RAMPs).
Evidence (as described)
- Human trial evidence exists, including a phase 2 trial and the later REDEFINE 1 trial. Trial results do not make an online research vial an approved medicine.
- A result reported with CagriSema cannot be attributed to cagrilintide alone, and a self-mixed combination is described as not automatically equivalent to the trial formulation.
